| In vitro | Methacycline remt TGF-β1-geïnduceerd α-gladde spier actine, Snail1 en collageen I van primaire alveolaire epitheelcellen. Methacycline remt TGF-β1-geïnduceerde niet-Smad-routes, inclusief c-Jun N-terminale kinase, p38 en Akt-activering, maar niet Smad of β-catenine transcriptionele activiteit. Methacycline heeft geen effect op baseline c-Jun N-terminale kinase, p38 of Akt-activiteiten of longfibroblastreacties op TGF-β1. | |
| In Vivo | Methacycline (100 mg/kg, i.p.) dagelijks toegediend vanaf 10 dagen na intratracheale bleomycine verbetert de overleving op dag 17. Methacycline vermindert bleomycine-geïnduceerde canonieke EMT-markers, Snail1, Twist1, collageen I, evenals fibronectine-eiwit en mRNA. Methacycline vermindert de accumulatie van ontstekingscellen niet en verandert TGF-β1-responsieve genen in alveolaire macrofagen niet. |
