Biologische activiteit

Beschrijving CH-223191 is een potente en specifieke aryl hydrocarbon receptor (AhR) antagonist met een IC50 van 30 nM.

CH-223191 blokkeert TCDD-gemedieerde nucleaire translocatie en DNA-binding van AhR, en veroorzaakt ook de remming van TCDD-ge orienteeerde cytochroom P450 enzymactiviteit.

In menselijke glioblastoomcellen downreguleert deze verbinding de TGF-beta/Smad-route en vermindert ze de clonogene overleving en invasiviteit.

In SK-N-SH menselijke neuronale cellen, gaat deze chemische stof de TCDD-ge orienteeerde onderdrukking van neuronale acetylcholinesterase-expressie tegen.

In endotheelcellen potentieert het de ICAM-1-expressie en voorkomt het de nucleaire translocatie van RelB.

In Vivo

CH-223191 (10 mg/kg/dag, p.o.) voorkomt krachtig TCDD-veroorzaakte cytochroom P450-inductie, levert rtoxiciteit en wasting-syndroom bij muizen.
