AT7519 ist ein ATP-kompetitiver CDK-Inhibitor mit einem Ki-Wert von 38 nM für CDK1. Diese Verbindung ist inaktiv gegenüber allen Nicht-CDK-Kinasen mit Ausnahme von GSK3β (IC50 = 89 nM). Sie zeigt eine potente antiproliferative Aktivität in einer Vielzahl menschlicher Tumorzelllinien mit IC50-Werten von 40 nM für MCF-7 bis 940 nM für SW620, was mit der Hemmung von CDK1 und CDK2 übereinstimmt. Diese Chemikalie induziert dosisabhängige Zytotoxizität in Myelom (MM)-Zelllinien mit IC50-Werten von 0,5 bis 2 μM nach 48 Stunden, wobei die empfindlichsten Zelllinien MM.1S (0,5 μM) und U266 (0,5 μM) und die resistenteste MM.1R (>2 μM) sind. Sie induziert keine Zytotoxizität in peripheren mononukleären Blutzellen (PBMNC). Dieser Inhibitor überwindet teilweise den proliferativen Vorteil, der durch IL6 und IGF-1 verliehen wird, sowie die schützende Wirkung von Knochenmark-Stromazellen (BMSCs). Er induziert eine schnelle Dephosphorylierung von RNA pol II CTD an den Serin 2- und Serin 5-Stellen und führt zur Hemmung der Transkription, was teilweise zu seiner induzierten Zytotoxizität von MM-Zellen beiträgt. Dieses Mittel induziert die Aktivierung von GSK-3β durch Herunterregulierung der GSK-3β-Phosphorylierung, was ebenfalls zu seiner induzierten apoptosis unabhängig von der Hemmung der Transkription beiträgt.

Eine zweimal tägliche Dosis von AT7519 (9,1 mg/kg) führt zu einer Tumorregression sowohl bei frühen als auch bei fortgeschrittenen s.c.-Tumoren in den HCT116- und HT29-Kolonkarzinom-Xenograftmodellen. Diese Verbindung (15 mg/kg) hemmt das Tumorwachstum und verlängert das mittlere Gesamtüberleben von Mäusen im humanen MM-Xenograft-Mausmodell in Verbindung mit einer erhöhten Caspase-3-Aktivierung.
