Beschreibung Dihydromyricetin (Ampelopsin, Ampeloptin) ist ein natürliches antioxidatives Flavonoid aus Ampelopsis grossedentata.
Ziele
GABAA receptor
In vitro Dihydromyricetin zeigt antioxidative Aktivität als effektiver Fänger des 1,1-Diphenyl-2-Picrylhydrazyl-Radikals (DPPH). Es hemmt auch den Anstieg der Lipidperoxidationswerte (LPO) im Linolsäuresystem, das durch FeSO4-Edetinsäure katalysiert wird. DHM (1 μM) antagonisiert als neuartiges Medikament gegen Alkoholintoxikation sowohl die akute EtOH-induzierte Potenzierung von GABA(A)Rs als auch die EtOH-Exposition/-Entzugs-induzierte GABA(A)R-Plastizität und erhöht die GABA(A)R-alpha4-Untereinheitsexpression im Hippocampus und in kultivierten Neuronen.
In vivo Dihydromyricetin (1 mg/kg, i.p.) wirkt akuter Alkohol-(EtOH)-Intoxikation entgegen und auch Entzugserscheinungen bei Ratten, einschließlich Toleranz, erhöhter Angst und Anfälligkeit für Krampfanfälle. LD50: Mäuse 1,41 g/kg (i.p.)
