Biologische Aktivität

Beschreibung Tocilizumab (anti-IL-6R) ist ein humanisierter monoklonaler Antikörper, der an den Interleukin-6-Rezeptor bindet, MW: 148 KD.
Ziele

In vitro

Nach Zugabe von Tocilizumab in verschiedenen Konzentrationen zu HMO6- und CHME5-Zellen, die mit F3-CM oder mit IL6 stimuliert wurden, nehmen die Expressionsniveaus der entzündungshemmenden M2-Marker (CD206 und Arg1) sowie die Niveaus von phosphoryliertem JAK2 und STAT3 ab.
In vivo

Tocilizumab kann die JAK2/STAT3-Aktivierung blockieren und die Hirnmetastasierung (BM) von A549-F3-Zellen in vivo behindern.

Protokoll (nur als Referenz)

Zell-Assay:

  • HMO6- und CHME5-Zellen mit Kontrollmedium oder F3-CM allein oder F3-CM wurden mit Tocilizumab (2,5 μg/ml für 24 h) oder mit IL6 (200 ng/ml für 24 h) allein oder IL6-ergänzt mit Tocilizumab behandelt. Western-Blot-Analyse von p-JAK2, JAK2, p-STAT3 und STAT3 in HMO6- und CHME5-Zellen wurden durchgeführt.
Tierstudie:

  • Tiermodelle: Nacktmäuse mit Hirnmetastasenmodellen
