Dapagliflozin ist gegenüber hSGLT1 mit einer 1200-fachen IC50 nicht empfindlich. Diese Verbindung ist 32-mal wirksamer als Phlorizin gegen hSGLT2, aber 4-mal weniger wirksam als Phlorizin gegen hSGLT1. Sie ist hochselektiv gegenüber GLUT-Transportern und zeigt eine 8–9%ige Hemmung in proteinfreiem Puffer bei 20 µM und praktisch keine Hemmung in Anwesenheit von 4% bovinem Serumalbumin. Diese Chemikalie weist eine gute Permeabilität durch Caco-2-Zellmembranen auf und ist ein Substrat für P-Glykoprotein (P-gp), aber kein signifikanter P-gp-Inhibitor. Sie ist in Ratten-, Hunde-, Affen- und menschlichem Serum bei 10 µM stabil. Sie zeigt keine hemmenden Reaktionen oder Induktion von menschlichen P450-Enzymen. Die in vitro-Stoffwechselwege dieser Verbindung sind Glucuronidierung, Hydroxylierung und O-Deethylierung. |

Chinesische Hamster-Ovarialzellen (CHO), die humanes SGLT2 (hSGLT2) und humanes SGLT1 (hSGLT1) stabil exprimieren, werden für die Entwicklung von Transportassays unter Verwendung des selektiven SGLT-Substrats α-Methyl-D-Glucopyranosid (AMG) verwendet. Dapagliflozin wird auf seine Fähigkeit getestet, die Aufnahme von [14C]AMG in einem proteinfreien Puffer über eine Inkubationszeit von 2 Stunden zu hemmen. Die Dosis-Wirkungs-Kurve wird an ein empirisches Vier-Parameter-Modell angepasst, um die Inhibitorkonzentration bei halbmaximaler Reaktion (EC50) zu bestimmen. Proteinfreier Puffer wird verwendet, um die proteinarmen Bedingungen des glomerulären Filtrats zu simulieren, das die SGLT-Ziele auf der luminalen Oberfläche des proximalen Tubulus in der Niere umspült.

Dapagliflozin senkt den Blutzuckerspiegel nach einer oralen Dosis von 0,1 mg/kg bei hyperglykämischen Streptozotocin (STZ)-Ratten um 55%, was teilweise auf die metabolische Stabilität zurückzuführen ist, die durch die C-Glucosid-Bindung verliehen wird. Diese Verbindung zeigt ein günstiges Absorptions-, Verteilungs-, Metabolismus- und Ausscheidungsprofil (ADME) und ist oral bioverfügbar. Diese Chemikalie (1 mg/kg) verursacht bei normalen Ratten über 24 Stunden nach der Dosis eine signifikante dosisabhängige Glukosurie und eine Zunahme des Urinvolumens. Sie induziert eine Zunahme der Urin-Glukose- und Urinvolumenausscheidung 6 Stunden nach der Dosis bei Zucker-Diabetischen-Fett (ZDF)-Ratten. Dieser Wirkstoff senkt die Nüchtern- und postprandialen Glukosespiegel bei ZDF-Ratten selbst nach 2 Wochen Behandlung, ohne Anzeichen von Nieren- oder Lebertoxizität. Es reduziert signifikant die Entwicklung von Hyperglykämie mit gesenktem Blutzucker. Diese Verbindung könnte die Insulinsensitivität verbessern, die β-Zellmasse reduzieren und die Entwicklung einer eingeschränkten Pankreasfunktion verringern. |
