Der Lck HTRF Kinase-Assay beinhaltet die ATP-abhängige Phosphorylierung eines biotinylierten Substratpeptids von Gastrin in Anwesenheit oder Abwesenheit einer Inhibitorverbindung. Die Endkonzentration von Gastrin beträgt 1,2 μM. Die Endkonzentration von ATP beträgt 0,5 μM (Km app = 0,6 ± 0,1 μM), und die Endkonzentration von Lck (eine GST-Kinase-Domänenfusion (AA 225−509)) beträgt 250 pM. Die Pufferbedingungen sind wie folgt: 50 mM HEPES pH = 7,5, 50 mM NaCl, 20 mM MgCl2, 5 mM MnCl, 2 mM DTT, 0,05 % BSA. Der Assay wird mit 160 μL Nachweisreagenz abgeschreckt und gestoppt. Die Nachweisreagenzien sind wie folgt: Puffer aus 50 mM Tris, pH = 7,5, 100 mM NaCl, 3 mM EDTA, 0,05 % BSA, 0,1 % Tween20. Vor dem Ablesen wird Streptavidin-Allophycocyanin (SA-APC) in einer Endkonzentration von 0,0004 mg/mL im Assay zusammen mit europiummarkiertem Anti-Phosphotyrosin-Antikörper (Eu-anti-PY) in einer Endkonzentration von 0,025 nM hinzugefügt. Die Assay-Platte wird in einem Discovery-Fluoreszenzplattenleser mit einer Anregung bei 320 nm und einer Emission bei 615 und 655 nm abgelesen. Assays für andere Kinasen werden auf ähnliche Weise wie oben beschrieben durchgeführt, wobei die Konzentrationen von Enzym, Peptidsubstrat und ATP, die der Reaktion zugesetzt werden, je nach spezifischer Aktivität der Kinase und gemessenen Kms für die Substrate variieren.
