Torin1 hemmt die Phosphorylierung von mTORC1- und mTORC2-Substraten in Zellen bei Konzentrationen von 2 bzw. 10 nM. Darüber hinaus zeigt Torin1 eine 1000-fache Selektivität für mTOR gegenüber PI3K (EC50 = 1800 nM) und eine 100-fache Bindungsselektivität gegenüber 450 anderen Proteinkinasen. Torin1 verursacht einen Zellzyklusarrest über einen rapamycinresistenten Mechanismus, der auch unabhängig von mTORC2 ist. Torin1 stört mTORC1-abhängige Phänotypen vollständiger als Rapamycin. Rapamycinresistente Funktionen von mTORC1 sind für die Cap-abhängige Translation erforderlich. Zur Herstellung von löslichem mTORC1 werden HEK-293T-Zelllinien, die N-terminal FLAG-markiertes Raptor stabil exprimieren, unter Verwendung von vesikulärem Stomatitis-Virus G-pseudotypisiertem MSCV-Retrovirus generiert. Für mTORC2 werden ähnliche HeLa-Zellen, die N-terminal FLAG-markiertes Protor-1 stabil exprimieren, generiert. Beide Komplexe werden durch Lysieren von Zellen in 50 mm HEPES, pH 7,4, 10 mm Natriumpyrophosphat, 10 mm Natrium-β-glycerophosphat, 100 mm NaCl, 2 mm EDTA, 0,3 % CHAPS gereinigt. Die Zellen werden 30 min bei 4 °C lysiert, und die unlösliche Fraktion wird durch Mikrozentrifugation bei 13.000 U/min für 10 min entfernt. Die Überstände werden 1 h lang mit FLAG-M2-monoklonalem Antikörper-Agarose inkubiert und dann dreimal mit Lyse-Puffer und einmal mit Lyse-Puffer, der eine Endkonzentration von 0,5 M NaCl enthält, gewaschen. Gereinigtes mTORC1 wird mit 100 μg/mL 3×FLAG-Peptid in 50 mm HEPES, pH 7,4, 100 mm NaCl eluiert. Das Eluat kann aliquotiert und bei -80 °C gelagert werden. Die Substrate S6K1 und Akt1 werden gereinigt. Kinase-Assays werden 20 min bei 30 °C in einem Endvolumen von 20 μL durchgeführt, bestehend aus Kinase-Puffer (25 mm HEPES, pH 7,4, 50 mm KCl, 10 mm MgCl2, 500 μm ATP) und 150 ng inaktivem S6K1 oder Akt1 als Substrate. Die Reaktionen werden durch Zugabe von 80 μL Probenpuffer gestoppt und 5 min gekocht. Die Proben werden anschließend mittels SDS-PAGE und Immunoblotting analysiert.
