AP24534 hemmt potent natives Abl, AblT315I und andere klinisch wichtige Abl-Kinase-Domänenmutanten mit einer IC50 von 0,30 nM–2 nM. Diese Verbindung hemmt weder Mitglieder der Aurora-Kinasefamilie noch den Insulinrezeptor oder CDK2/Cyclin E. Sie hemmt die Proliferation von Ba/F3-Zellen, die Bcr-Abl exprimieren, mit einer IC50 von 0,5 nM, sowie Ba/F3-Zellen, die eine Reihe von Bcr-Abl-Mutanten exprimieren, mit einer IC50 von 0,5 nM–36 nM. Die Hemmung der Proliferation durch diese Chemikalie korreliert mit der Induktion von Apoptose. In Leukämiezelllinien, die aktivierte Formen von FLT3-, KIT-, FGFR1- und PDGFRα-Rezeptoren enthalten, hemmt es potent die Rezeptorphosphorylierung und die Zellproliferation mit einer IC50 von 0,3 nM bis 20 nM. In MV4-11 (FLT3-ITD(+/+))-, aber nicht in RS4;11 (FLT3-ITD(–/–))-AML-Zellen hemmt dieser Inhibitor die FLT3-Signalübertragung und induziert Apoptose bei weniger als 10 nM. Er hemmt die Lebensfähigkeit primärer Leukämie-Blasten von einem FLT3-ITD-positiven AML-Patienten mit einer IC50 von 4 nM, aber nicht die von Patienten mit AML, die natives FLT3 exprimieren. In Ba/F3-Zellen, die zur Expression von aktiviertem FGFR1-4 entwickelt wurden, hemmt dieses Mittel potent die FGFR-vermittelte Signalübertragung und Lebensfähigkeit mit einer IC50 von weniger als 40 nM. Die Wirkung von AP24534 (0-320 nM) auf die GST-Abl-Kinaseaktivität wird unter Verwendung eines synthetischen Peptidsubstrats (Abltide: EAIYAAPFAKKK) bewertet. Die Assays werden bei 30 °C für 15 Minuten in einem 25 μL Reaktionsgemisch durchgeführt: 8 mM MOPS (pH 7), 0,2 mM EDTA, 50 μM Abltide, 30 mM MgCl2, 10 mM β-Glycerophosphat, 1 mM EGTA, 0,002% Brij-35, 0,4 mM DTT, 0,2 mg/mL BSA, 0,4 mM Natriumorthovanadat, 10 nM WT oder mutierte GST-Abl-Kinase und 100 µM ATP/γ-32[P]ATP (5000 cpm/pmol). Die Reaktionen werden durch Übertragung eines Teils des Reaktionsgemisches auf einen p81 Phosphocellulosefilter und Eintauchen in 0,75%ige Phosphorsäure beendet. Die Filter werden 3-mal in 0,75%iger Phosphorsäure gewaschen, in Aceton gespült und an der Luft getrocknet; der Phosphateinbau wird durch Szintillationszählung bestimmt. Alle Ergebnisse werden für die Hintergrundbindung an die Filter korrigiert, die durch Weglassen des Peptidsubstrats aus der Kinase-Reaktion bestimmt wird. Zeitverlaufsexperimente zur Bestimmung des linearen Bereichs der enzymatischen Aktivität gehen den Kinase-Assays voraus.

In einem Maus-Xenograft-Modell von Ba/F3-Zellen, die natives Bcr-Abl exprimieren, verlängert diese Verbindung (2,5 mg/kg und 5 mg/kg) die mittlere Überlebenszeit der Mäuse. Im Xenograft-Modell von Ba/F3 Bcr-AblT315I unterdrückt diese Verbindung (10 mg/kg–50 mg/kg) signifikant das Tumorwachstum. Sie (30 mg/kg) verringert die phosphorylierten Bcr-Abl- und phosphorylierten CrkL-Spiegel in den Tumoren.

Our 10mM solution in DMSO is in pure DMSO and there is not other stabilizer added. You can use it for animal study. The vehicle we suggest for this compound is: 30% PEG400/0.5% Tween80/5% Propylene glycol. It is a milky suspension used for oral gavage.
