In vitro

Pazopanib hemmt die VEGF-induzierte Phosphorylierung von VEGFR2 in HUVEC-Zellen mit einer IC50 von 8 nM.

Diese Verbindung zeigt eine dosisabhängige Wachstumshemmung in allen synovialen Sarkomzelllinien, einschließlich SYO-1- und HS-SY-II-Zellen. Die Proliferation von SYO-1- und HS-SY-II-Zellen wird bereits bei 1 µg/mL dieser Chemikalie gehemmt und bei 5 µg/mL vollständig aufgehoben. Sie induziert einen G1-Arrest und unterdrückt dadurch das Wachstum von synovialen Sarkomzellen. Die Phosphorylierung von Akts, GSK-3β, JNKs, p70 S6 Kinase und mTOR wird in mit dieser Verbindung behandelten SYO-1-Zellen im Vergleich zu den mit dem Vehikel behandelten Zellen unterdrückt.

Diese Chemikalie zwischen 20 µg/mL und 22,5 µg/mL zeigt eine zunehmende Reduzierung der RPE-Zellviabilität.

Kinase-Assay
Kinase-Enzym-Assays
VEGFR-Enzym-Assays für VEGGR1, VEGFR2 und VEGFR3 werden im HTRF-Format (Homogeneous Time-Resolved Fluorescence) in 384-Well-Mikrotiterplatten unter Verwendung eines gereinigten, Baculovirus-exprimierten Glutathion-S-Transferase (GST)-Fusionsproteins, das den katalytischen c-Terminus der humanen VEGFR-Rezeptor-Kinasen 1, 2 oder 3 kodiert, durchgeführt. Die Reaktionen werden durch Zugabe von 10 μL aktivierter VEGFR2-Kinase-Lösung [Endkonzentration, 1 nM Enzym in 0,1 M HEPES, pH 7,5, enthaltend 0,1 mg/mL Rinderserumalbumin (BSA), 300 μM Dithiothreitol (DTT)] zu 10 μL Substratlösung [Endkonzentration, 360 nM Peptid, (Biotin-Aminohexyl-EEEEYFELVAKKKK-NH2), 75 μM ATP, 10 μM MgCl2] und 1 μL titrierter Verbindung in DMSO initiiert. Die Platten werden 60 Minuten bei Raumtemperatur inkubiert, und dann wird die Reaktion durch Zugabe von 20 μL 100 mM Ethylendiamintetraacetat (EDTA) gestoppt. Nach dem Stoppen werden 20 μL HTRF-Reagenzien (Endkonzentration, 15 nM Streptavidin-gebundenes Allophycocyanin, 1 nM Europium-markierter Anti-Phosphotyrosin-Antikörper, verdünnt in 0,1 mg/mL BSA, 0,1 M HEPES, pH 7,5) hinzugefügt und die Platten für mindestens 10 Minuten inkubiert. Die Fluoreszenz bei 665 nM wird mit einem Wallac Victor Plattenlesegerät unter Verwendung einer Zeitverzögerung von 50 μs gemessen.
In vivo

Die mit 30 mg/kg oder 100 mg/kg Pazopanib behandelten Mäuse zeigen eine signifikante Abnahme der Tumorlast im Vergleich zu den mit Vehikel oder 10 mg/kg dieser Verbindung behandelten Mäusen. Die Behandlung mit diesem Mittel wird gut vertragen und es gibt keinen signifikanten Unterschied im Körpergewicht zwischen den Mäusen in jeder Gruppe.
